Annotation

The article presents the results of assessing the physiological state of new self-rooted hybrid forms of table grape Akello, Artek, Valencia and Courage growing in the Krasnodar Territory in the summer period. The cultivar Livia of Russian selection was selected as a control. Artificial drought was simulated in a laboratory conditionS. After stress effect, the following physiological parameters of grape leaves were analyzed the degree of damage to cell membranes (based on the content of malondialdehyde, one of the products of lipid peroxidation of cell membranes), the relative water content, the quantum yield of the photochemical reaction of photosystem II, the content of chlorophyll a and the ratio of a/b chlorophylls. It was found that the most depressed month for the studied grape plants was July. This is evidenced by the averaged indicators of studied grape plants according to the analyzed parameters low values of photosynthesis and relative water content in the leaves and increased level of malondialdehyde, which determines the intensity of the development of oxidative processes by the influence of negative factors. The greatest adaptive potential in this month was revealed in the Artek hybrid form, which had high value of photosynthesis and a low level of malondialdehyde content. In August, the hybrid forms of Artek and Courage were characterized by high values of the relative water content in the leaves and low value of malondialdehyde, and besides the level of chlorophyll a fluorescence (quantum yield) did not change compared to July. The hybrid Valencia form had the highest value of malondialdehyde and the lowest value of relative water content during the summer period, which makes it the least resistant form among the studied. The hybrid form of Artek was identified as the most resistant form to influence of summer stress factors in the territory of Krasnodar region. Further, this form can be recommend for the creation of stable ampelocenoses.

Annotation

Domestic varieties resistant to abiotic and biotic stressors with high profitability in production are needed to replace the introduced table grape varieties. The aim of this research was to study promising hybrid forms of grapes in grafted and own-rooted culture in the conditions of the Central agroecological zone of viticulture of the Krasnodar region. The objects were hybrids of grapes Vitis L. Agat Dubovskiy, Akello, Ispolin, Kishmish Dubovskiy. The control variety was Livia. Rootstock Berlandieri x Riparia SO4. The planting scheme of the bushes was 4.0 × 2.0 m, the formation of bushes is a high-standard two-armed cordon, drip irrigation. The climate is moderately continental, the average annual air temperature is +12.5 ...+13.0 ºC, the maximum reaches +40 ºC, the absolute minimum temperature is -28...-30 ºC. The sum of active air temperatures is equal to 3900 ...4100 ºC. The total precipitation is 700-800 mm. The studies were carried out using generally accepted field, laboratory and statistical methods. According to phenological observations, variety and hybrids were distinguished by three maturation periods according to the international classification very early Livia (112 days) and Akello (114 days), medium Agat Dubovskiy (142 days) and Ispolin (145 days) and medium-late Kishmish Dubovskiy (148 days). The phenology of grafted and own-rooted plants differed slightly. Livia variety and hybrids required from 2467 ºC to 3243 ºC for vegetation. According to agrobiological indicators the number of fruiting shoots, inflorescences, fruitfulness coefficients grafted hybrid forms are more productive than the control variety and their own-rooted analogues, with the exception of Kishmish Dubovskiy, whose fruitfulness Coefficients are similar in the grafted and own-rooted culture and are close to the control values. The relative water content (RWC) of grafted and own-rooted plants is above 80 %, which means a normally hydrated leaf.

Annotation

This work provides a comparison results of the physiological and biochemical characteristics of two apple cultivars different ploidy Soyuz (2n=3x) and Prikubanskoe (2n=2x). We studied the content of protein, lignin, malondialdehyde (MDA) and the level of peroxidase activity in annual shoots and leaves, as well as the content of nucleic acids in mature seeds. We found that the triploid of Soyuz is characterized by higher average values for almost parameters studied and their lower variability unlike the diploid of Prikubanskoe. At the same time, statistically significant differences were obtained by the content of protein and nucleic acids in the shoots and the seeds, respectively. The Soyuz variety, as a triploid, had higher values of these characteristics. On the other hand, Soyuz apple variety had the lower degree of resistance to winter temperatures compared to Prikubanskoe. It was determined by the lower activity of peroxidase, one of the antioxidant enzymes protector of plants under stress conditions and by the higher content of MDA, which is a product of lipid peroxidation of cell membranes. Our studies showed that the Soyuz apple variety, despite the relative stability of physiological characteristics in the summer months of study have, had the low level of stress tolerance compared to the Prikubanskoe variety in the winter period.

Annotation

At present, the adaptive stability potential of some fruit varieties is insufficient for a response to climate changes in the North Caucasus Region of Russia. In this regard, the creation of varieties with increased environmental plasticity and high adaptive potential is relevant. Physiological and biochemical studies of apple varieties state after artificially induced low-temperature stress will allow to assess the studied apple varieties for resistance to critical frosts in mid-winter. A comparative assessment of the apple variety resistance to low temperature stress showed that there were quantitative changes content of malon dialdehyde content in the one-year shoots, as well as ascorbic acid, anthocyanins, chalcones, and peroxidase activity. It is established that in various apple varieties, peroxidase activity and antioxidant content in the tissue of shoots under conditions of low temperature stress varied with difference, detoxification of active oxygen forms (AOF) was achieved by various components of antioxidant protection. In the Prikubanskoye apple variety, the content of malondialdehyde in the one-year shoots remained almost unchanged, indicating the greater stability of the cell membranes, achieved due to the increased activity of peroxidase. n the Rodnichok variety, AOF detoxification is achieved by increasing in the ascorbic acid content by 3 times. Varieties of Rassvet and Prikubanskoe had the highest content of anthocyanins and chalcones, that indicate their greater frost resistance in the middle of winter in comparison with other studied varieties. Analysis of the physiological and biochemical characteristics we have found as a result of our research showed that Prikubanskoe apple variety is resistant (II component of winter hardiness). The varieties of Rassvet, Rodnichok, Idared are of less resistant to the highest frost in the mid-winter.

Annotation

Micropropagation is a promising direction for obtaining a healthy, genetically homogeneous grape planting material. The efficiency of reproduction is determined by many factors, including the acclimatization of plants to natural environmental conditions. The difficulty of transferring in vitro plants to non-sterile conditions is associated with a number of physiological and anatomical features of in vitro plants, among which the low level of photosynthetic processes is of particular importance. The influence of various concentrations of sucrose in the culture medium and additives, simulating environmental stresses on the rates of photosynthesis of grapes during acclimatization, was studied. The content of sucrose 10g/l in the culture medium was the most optimal for the formation of the photosynthetic apparatus. Both low and high sucrose content in the culture medium negatively affected the formation of pigments and the efficiency of photosynthetic processes. The introduction of PEG and NaCl into the nutrient medium led to a decrease in the total content of pigments, the quantum yield of photochemical reactions, but increased the expression of RUBISCO activase. The use of ABA as part of the culture medium had a positive effect on the formation of pigments in grape plants during acclimatization. The effects of weak stresses on the photosynthetic parameters of grapes in in vitro culture and during the acclimatization period are identical to the effects of drought and salinity under natural conditions. At the same time, these types of influence have a positive effect on adaptive anti-stress reactions that prevent plant damage under ex vitro conditions, which ultimately increases the level of growth processes and the yield of seedlings during micropropagation of grapes.
