Annotation

The method of clonal plants micro reproduction in vitro is one of basic elements of technology of production of the crop's revitalized planding material. It is known that for each new variety it is required the carrying out all of elements of improvement technique: the selection of optimal ratios of elements of nutrient mediums (micro and macro elements, vitamins, growth substances, etc.), the determination of optimal terms of in vitro introduction, the search of safe and effective sterilizers. In the horticulture of the South of Russia there is an annual assortment replenishment of new fruit varieties, so it is required the modification of technique for cultivation of these varieties. The purpose of our research is selection of the effective sterilizing agent for introduction in vitro of plum varieties. The objects of research are the preparations used for the surface disinfection of the explants in the process of introduction in vitro. The carried out research have shown that the greatest exit of viable explants is noted when processing by mercury iodide an average 82 %. Death of explants from an infection in this option is 8 % (the lowest among the studied options), a necrosis of explants an average 10 %, it is also the one of the lowest indexes in an experiment. The highest level of an infection is determined in a control option, when washing of plum explants by the distilled water an average 72 %. In the option of processing by the distilled water (control) the necrosis of apexes is an average 10 % that testifies of weak phytotoxic infection of this way of explants preparing in the process of plum reproduction.

Annotation

In this paper the results of work on studying and improvement of cultivation technology of landing material of plum and other stone fruit crops, free from bacterial crown gall are presented. The relevance of research is defined by great economic value of bacterial crown gall in the process of stone fruit crops cultivation. rown gall is the bacterial root disease of fruit and berry plants which is appeared in the form of tumors formation on roots and at root neck. Now the active expansion of the decease area is observed. Crown gall causes the greatest damage in the nurseries and at cultivation of trees in the young garden. On the basis of these own research and the analysis of references the system of measures including several stages is developed: the choice and preparation of the plot with carrying out of the bacteriological analysis and agrochemical inspection of the plot's soil, the preparation and sanitation of landing material, the laying of mother plantation, for its the works maintenance, the laying of fields for reproduction, the quarantine. During vegetation it is necessary to carry out monthly expeditious monitoring of phytosanitary condition of uterine plants and annual diagnostics of plants and soils regarding settling of Agrobacterium tumefaciens. The period of operation of uterine plantings, free from root cancer not more than 12 years. These methodical approaches are for industrial technologies of nurseries improvement from bacterial cancer of roots, and they establish the general rules to the choice and preparation of the plot for laying of nursery, the ways and methods of maintenance of soils in the nursery, according to requirements to the grown-up landing and uterine material. These rules are intended for specialists of the specialized enterprises, which are engaged in reproduction of landing material for stone fruit crops.

Annotation

The method of embryo culture has more and more wide application in the interspecific hybridization of fruit plants. For its improvement the hormonal regulation of growth and development of embryos in vitro is researched and the optimal age of embryos injected into culture is defined. The main objective of the presented work is to receive the viable plants from puny seeds and not sprout embryos of hybrids of sweet cherry and cherry in the course of their cultivation in vitro and to allocate the effective growth regulators and to establish the optimal terms of embryos input in the culture. The results of research on cultivation in vitro of sweet cherry and cherry of 16 combinations of crossing are presented. The regenerants of hybrid embryos of cherry and sweet cherry in vitro culture from 16 combinations of crossings are received. For embryos culture of sweet cherry and cherry the best modification of Murashige-Skoog environment with 79 % level of embryos regeneration is allocated. The most effective concentration of gibberellic acid in the environment for cultivation of cherry and sweet cherry is revealed. That is 10 mg of gibberellic acid per 1 liter of the Murashige-Skoog environment, for all that the embryo size should be maximized.

Annotation

Apple-tree is the leader in the harvesting and square of orchards in the horticulture of Russia. The development of protection systems from harmful organisms, as a way to increase in yield, is widely studied and put into practice of horticulture. At the same time, the morphological and physiological features of generative capacity and bearing in connection with pathogenesis of major apple diseases were not investigated enough. The aim of this research is to learn an apple organogenesis in the connection with the pathophytocenosis dynamics (for example, scab and powdery mildew of apple). The basic method of study is the morphological-physiological analysis, which combines the study of intra-buds development and the morphometry of the extra-buds development of shoots, leaves, flowers, ovary and fruits in the connection with the pathogenesis of the studied apple tree diseases. The results of studying of apple-tree organogenesis and analysis of the special features of the pathogenesis of the apple scab (Venturia inaequalis) and apple powdery mildew (Podosphaera leucotricha (Ell. et Ev.) Salm.), show that both processes depend on the climatic conditions in the location of agrical phytopathocenosis. The mechanism of regulation of apple tree organogenesis and development of pathophytocenosis of cultivation zone of orchards, are the climatic conditions (temperature regime, duration of day, amount of precipitation and others) and the degree of joining is sufficiently close. As consequence, the analysis of the process of apple-tree generative development makes possible to determine not only the data of the harvesting, the methods and the degree of pruning, the norm and the regimes of irrigation, and the introduction of fertilizers, but also the norms and the regimes of the pesticides application, which can be corrected taking into account the data of morphological-physiological analysis.
